NM_017636.4(TRPM4):c.3181C>T (p.Gln1061Ter) was classified as Uncertain significance by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: TRPM4 c.3181C>T (p.Gln1061X) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, however current evidence is not sufficient to establish loss of function as a mechanism for disease. The variant was absent in 251458 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.3181C>T has been observed in a genetic testing for Cardiomyopathy without strong evidence for causality (Lesurf_2022). These report(s) do not provide unequivocal conclusions about association of the variant with Progressive Familial Heart Block Type 1B. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication have been ascertained in the context of this evaluation (PMID: 35288587). No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as uncertain significance.
