This variant lies in the CYP21A2 gene (transcript NM_000500.9) at coding-DNA position 905, where C is replaced by A; at the protein level this means replaces serine at residue 302 with tyrosine — a missense variant. Submitter rationale: Variant summary: CYP21A2 c.905C>A (p.Ser302Tyr) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change. The variant was absent in 246420 control chromosomes (gnomAD). c.905C>A has been observed in individuals affected with Congenital Adrenal Hyperplasia/Non-Classic (e.g. Stikkelbroeck_2003, Stroek_2021). These data indicate that the variant is likely to be associated with disease. At least one publication reports experimental evidence evaluating an impact on protein function.
