This variant lies in the CFTR gene (transcript NM_000492.4) at 3 bases into the intron immediately after coding-DNA position 1679, where G is replaced by A. Submitter rationale: Variant summary: CFTR c.1679+3G>A alters a conserved nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 250328 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1679+3G>A has been observed in at least two individuals affected with Cystic Fibrosis (example: Terzic_2019). These reports do not provide unequivocal conclusions about association of the variant with Cystic Fibrosis. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
