NM_000203.5(IDUA):c.1373T>C (p.Leu458Pro) was classified as Likely pathogenic for Mucopolysaccharidosis type 1 by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: IDUA c.1373T>C (p.Leu458Pro) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change. The variant was absent in 21918 control chromosomes. c.1373T>C has been observed in multiple homozygous individuals of Indian descent affected with clinical features and/or diagnosis of Mucopolysaccharidosis Type 1 (Thomas_2021), including at least 1 family where it segregated with disease. These data indicate that the variant is very likely to be associated with disease, however the possibility of a subpopulation-specific common polymorphism cannot be entirely ruled out. While patient data suggest decreased enzymatic activity, the quality of the methods/assays could not be validated (example, Thomas_2021).
