This variant lies in the GALT gene (transcript NM_000155.4) at coding-DNA position 260, where C is replaced by T; at the protein level this means replaces proline at residue 87 with leucine — a missense variant. Submitter rationale: Variant summary: GALT c.260C>T (p.Pro87Leu) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 251490 control chromosomes. c.260C>T has been observed in at-least two individuals affected with Galactosemia (Jezela-Stanek_2021). These data indicate that the variant may be associated with disease. A different variant affecting the same codon has been classified as pathogenic by our lab (c.260C>G, p.Pro87Arg), supporting the critical relevance of codon 87 to GALT protein function. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
