Pathogenic for Mitochondrial DNA depletion syndrome 1 — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001953.5(TYMP):c.877T>C (p.Cys293Arg), citing LabCorp Variant Classification Summary - May 2015: Variant summary: TYMP c.877T>C (p.Cys293Arg) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The frequency data for this variant in gnomAD is considered unreliable, as metrics indicate poor data quality at this position. c.877T>C has been observed in multiple related individuals affected with Mitochondrial DNA Depletion Syndrome 1 (MNGIE type) from a large consanguineous pedigree (Redha_2024). These data indicate that the variant is very likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
