Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005732.4(RAD50):c.1718A>C (p.Lys573Thr), citing Ambry Variant Classification Scheme 2023: The p.K573T variant (also known as c.1718A>C), located in coding exon 11 of the RAD50 gene, results from an A to C substitution at nucleotide position 1718. The lysine at codon 573 is replaced by threonine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
