NM_005732.4(RAD50):c.3511G>A (p.Glu1171Lys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E1171K variant (also known as c.3511G>A), located in coding exon 23 of the RAD50 gene, results from a G to A substitution at nucleotide position 3511. The glutamic acid at codon 1171 is replaced by lysine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
