Submitter rationale: ABCB11 c.-24C>A is a variant located in the 5′ untranslated region (5′ UTR). This variant has been observed in at least one proband with an ABCB11-related disorder (PMID:28150711). At least one splicing study demonstrated that this variant results in aberrant splicing (PMID:28150711). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify ABCB11 c.-24C>A as a likely pathogenic variant.
