NM_000443.4(ABCB4):c.3226A>T (p.Ser1076Cys) was classified as Uncertain significance for Familial intrahepatic cholestasis by Genomenon, Inc, Genomenon, Inc, citing Genomenon Sequence Variant Interpretation Standards - Updated. This variant lies in the ABCB4 gene (transcript NM_000443.4) at coding-DNA position 3226, where A is replaced by T; at the protein level this means replaces serine at residue 1076 with cysteine — a missense variant. Submitter rationale: ABCB4 p.Ser1076Cys (c.3226A>T) is a missense variant that changes the amino acid at residue 1076 from Serine to Cysteine. This variant has been observed in at least one proband with an ABCB4-related disorder (PMID:37701337). At least one functional study has demonstrated a substantial alteration in protein function relative to the wild-type (PMID:28012258). It is absent or not present at a significant frequency in gnomAD. In silico models predict that this variant is possibly or probably damaging. In conclusion, we classify ABCB4 p.Ser1076Cys (c.3226A>T) as a variant of uncertain significance.
