Pathogenic for Familial intrahepatic cholestasis — the classification assigned by Genomenon, Inc, Genomenon, Inc to NM_000443.4(ABCB4):c.3100dup (p.Ile1034fs), citing Genomenon Sequence Variant Interpretation Standards - Updated. This variant lies in the ABCB4 gene (transcript NM_000443.4) at coding-DNA position 3100, duplicating one base; at the protein level this means shifts the reading frame starting at isoleucine residue 1034, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: ABCB4 p.Ile1034AsnfsTer4 (c.3100dup) is a frameshift variant that results in the production of a truncated protein which is predicted to undergo nonsense-mediated mRNA decay. This variant has been observed in at least one proband with an ABCB4-related disorder (PMID:36550572). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify ABCB4 p.Ile1034AsnfsTer4 (c.3100dup) as a pathogenic variant.
