This variant lies in the ATP8B1 gene (transcript NM_001374385.1) at the canonical splice acceptor site of the intron immediately before coding-DNA position 3401, where A is replaced by G; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: ATP8B1 c.3401-2A>G is a canonical splice variant affecting the acceptor splice site of intron 26. It is predicted to affect mRNA splicing, leading to a deleterious effect on the ATP8B1 protein. This variant has been observed in at least one proband with features of ATP8B1-deficiency (PMID:26382629). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify ATP8B1 c.3401-2A>G as a pathogenic variant.
