Uncertain significance for Familial intrahepatic cholestasis — the classification assigned by Genomenon, Inc, Genomenon, Inc to NM_000443.4(ABCB4):c.1691A>G (p.Asp564Gly), citing Genomenon Sequence Variant Interpretation Standards - Updated. This variant lies in the ABCB4 gene (transcript NM_000443.4) at coding-DNA position 1691, where A is replaced by G; at the protein level this means replaces aspartic acid at residue 564 with glycine — a missense variant. Submitter rationale: ABCB4 p.Asp564Gly (c.1691A>G) is a missense variant that changes the amino acid at residue 564 from Aspartic acid to Glycine. This variant has been observed in at least one proband with an ABCB4-related disorder (PMID:11313315;37701337). At least one functional study has demonstrated a substantial alteration in protein function relative to the wild-type (PMID:36674751). It is absent or not present at a significant frequency in gnomAD. In silico models predict that this variant is possibly or probably damaging. In conclusion, we classify ABCB4 p.Asp564Gly (c.1691A>G) as a variant of uncertain significance.
