This variant lies in the ABCB4 gene (transcript NM_000443.4) at coding-DNA position 1375, where G is replaced by C; at the protein level this means replaces aspartic acid at residue 459 with histidine — a missense variant. Submitter rationale: ABCB4 p.Asp459His (c.1375G>C) is a missense variant that changes the amino acid at residue 459 from Aspartic acid to Histidine. This variant has been observed in at least one proband with an ABCB4-related disorder (PMID:24594635). At least one functional study has demonstrated a substantial alteration in protein function relative to the wild-type (PMID:24594635;26900700). It is absent or not present at a significant frequency in gnomAD. In silico models predict that this variant is possibly or probably damaging. In conclusion, we classify ABCB4 p.Asp459His (c.1375G>C) as a variant of uncertain significance.
