This variant lies in the ABCB4 gene (transcript NM_000443.4) at the canonical splice donor site of the intron immediately after coding-DNA position 80, where G is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: ABCB4 c.80+1G>C is a canonical splice variant affecting the donor splice site of intron 2. It is predicted to affect mRNA splicing, leading to a deleterious effect on the ABCB4 protein. This variant has been observed in at least one proband with an ABCB4-related disorder (PMID:36550572). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify ABCB4 c.80+1G>C as a pathogenic variant.
