It is predicted to affect mRNA splicing, leading to a deleterious effect on the ABCB4 protein. This variant has been observed in at least one proband with an ABCB4-related disorder (PMID:12713886). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify ABCB4 c.344+2T>C as a pathogenic variant.
