Pathogenic for Familial intrahepatic cholestasis — the classification assigned by Genomenon, Inc, Genomenon, Inc to NM_001374385.1(ATP8B1):c.1227del (p.Glu409_Val410insTer), citing Genomenon Sequence Variant Interpretation Standards - Updated. This variant lies in the ATP8B1 gene (transcript NM_001374385.1) at coding-DNA position 1227, deleting one base. Submitter rationale: ATP8B1 p.Val410Ter (c.1227del) is a nonsense variant that introduces a premature stop codon at amino acid position 410, creating a truncated protein that is predicted to undergo nonsense-mediated mRNA decay. This variant has been observed in at least one proband with features of ATP8B1-deficiency (PMID:22525741). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify ATP8B1 p.Val410Ter (c.1227del) as a pathogenic variant.
