This variant lies in the PTEN gene (transcript NM_000314.8) at coding-DNA position 356, where T is replaced by G; at the protein level this means replaces valine at residue 119 with glycine — a missense variant. Submitter rationale: The p.V119G variant (also known as c.356T>G), located in coding exon 5 of the PTEN gene, results from a T to G substitution at nucleotide position 356. The valine at codon 119 is replaced by glycine, an amino acid with dissimilar properties. In a massively parallel functional assay using a humanized yeast model, lipid phosphatase activity for this variant was functionally deficient (Mighell TL et al. Am J Hum Genet, 2018 May;102:943-955). This variant also demonstrated low intracellular protein abundance in a massively parallel functional assay (Matreyek KA et al. Nat Genet, 2018 Jun;50:874-882).
