Pathogenic for Familial intrahepatic cholestasis — the classification assigned by Genomenon, Inc, Genomenon, Inc to NM_003742.4(ABCB11):c.11C>G (p.Ser4Ter), citing Genomenon Sequence Variant Interpretation Standards - Updated: ABCB11 p.Ser4Ter (c.11C>G) is a nonsense variant that introduces a premature stop codon at amino acid position 4, creating a truncated protein that may be subject to nonsense-mediated mRNA decay. This variant has been observed in at least one proband with an ABCB11-related disorder (PMID:28730136). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify ABCB11 p.Ser4Ter (c.11C>G) as a pathogenic variant.
