This variant lies in the SPINK5 gene (transcript NM_006846.4) at 17 bases into the intron immediately before coding-DNA position 411, where C is replaced by G. Submitter rationale: A novel splicing variant, g.148088525C>G (NM_006846.4:c.411-17C>G) in intron 5 of SPINK5 gene was identified in homozygous state in the proband. This variant is absent in homozygous state and/or heterozygous state in gnomAD (v4.1.0) population database and our in-house exome data of 3999 individuals. SpliceAI tool has predicted this variant to result in aberrant splicing which may lead to either the formation of a truncated protein product or the transcript to undergo nonsense mediated mRNA decay.
