NM_001005242.3(PKP2):c.643G>A (p.Asp215Asn) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D215N variant (also known as c.643G>A), located in coding exon 3 of the PKP2 gene, results from a G to A substitution at nucleotide position 643. The aspartic acid at codon 215 is replaced by asparagine, an amino acid with highly similar properties. This variant has been detected in an individual with idiopathic ventricular fibrillation (Visser M et al. Heart Rhythm, 2017 Jul;14:1035-1040). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
