Uncertain significance — the classification assigned by Ambry Genetics to NM_006648.4(WNK2):c.5888T>G (p.Leu1963Arg), citing Ambry Variant Classification Scheme 2023: The p.L1963R variant (also known as c.5888T>G), located in coding exon 23 of the WNK2 gene, results from a T to G substitution at nucleotide position 5888. The leucine at codon 1963 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
