Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006361.6(HOXB13):c.32G>A (p.Gly11Glu), citing Ambry Variant Classification Scheme 2023: The p.G11E variant (also known as c.32G>A), located in coding exon 1 of the HOXB13 gene, results from a G to A substitution at nucleotide position 32. The glycine at codon 11 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
