NM_020937.4(FANCM):c.386A>G (p.Tyr129Cys) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y129C variant (also known as c.386A>G), located in coding exon 1 of the FANCM gene, results from an A to G substitution at nucleotide position 386. The tyrosine at codon 129 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
