The aspartic acid at codon 224 is replaced by asparagine, an amino acid with highly similar properties. This variant was reported in individual(s) with hypertrophic cardiomyopathy, though patient-specific details were limited (Harper AR et al. Nat Genet, 2021 Feb;53:135-142; Ambry internal data). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive.
