Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001277115.2(DNAH11):c.12707A>T (p.Asn4236Ile), citing Ambry Variant Classification Scheme 2023: The p.N4236I variant (also known as c.12707A>T), located in coding exon 77 of the DNAH11 gene, results from an A to T substitution at nucleotide position 12707. The asparagine at codon 4236 is replaced by isoleucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
