Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_002691.4(POLD1):c.2954-2del, citing Ambry Variant Classification Scheme 2023: The c.2954-2delA intronic variant, located in intron 22 of the POLD1 gene, results from a deletion of one nucleotide within intron 22 of the POLD1 gene. This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice acceptor site. Variants that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay. However, loss of function has not been established as a mechanism of disease. Based on the available evidence, the clinical significance of this variant remains unclear.
