Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_003073.5(SMARCB1):c.372G>T (p.Lys124Asn), citing Ambry Variant Classification Scheme 2023: The p.K124N variant (also known as c.372G>T), located in coding exon 4 of the SMARCB1 gene, results from a G to T substitution at nucleotide position 372. The lysine at codon 124 is replaced by asparagine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
