Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000057.4(BLM):c.3863G>C (p.Trp1288Ser), citing Ambry Variant Classification Scheme 2023: The p.W1288S variant (also known as c.3863G>C), located in coding exon 19 of the BLM gene, results from a G to C substitution at nucleotide position 3863. The tryptophan at codon 1288 is replaced by serine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
