Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_003238.6(TGFB2):c.881A>G (p.Gln294Arg), citing Ambry Variant Classification Scheme 2023: The p.Q294R variant (also known as c.881A>G), located in coding exon 5 of the TGFB2 gene, results from an A to G substitution at nucleotide position 881. The glutamine at codon 294 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
