Uncertain significance — the classification assigned by Ambry Genetics to NM_002386.4(MC1R):c.128G>A (p.Gly43Glu), citing Ambry Variant Classification Scheme 2023: The p.G43E variant (also known as c.128G>A), located in coding exon 1 of the MC1R gene, results from a G to A substitution at nucleotide position 128. The glycine at codon 43 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
