NM_000535.7(PMS2):c.18C>G (p.Ser6Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S6R variant (also known as c.18C>G), located in coding exon 1 of the PMS2 gene, results from a C to G substitution at nucleotide position 18. The serine at codon 6 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is poorly conserved on limited sequence alignment. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
