NM_004656.4(BAP1):c.849G>T (p.Glu283Asp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E283D variant (also known as c.849G>T), located in coding exon 10 of the BAP1 gene, results from a G to T substitution at nucleotide position 849. The glutamic acid at codon 283 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
