Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004656.4(BAP1):c.1870G>C (p.Gly624Arg), citing Ambry Variant Classification Scheme 2023: The p.G624R variant (also known as c.1870G>C), located in coding exon 14 of the BAP1 gene, results from a G to C substitution at nucleotide position 1870. The glycine at codon 624 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
