Pathogenic for Cardiovascular phenotype — the classification assigned by Ambry Genetics to Single allele, citing Ambry Variant Classification Scheme 2023: The c.3258delG variant, located in coding exon 2 of the ZNF469 gene, results from a deletion of one nucleotide at nucleotide position 3258, causing a translational frameshift with a predicted alternate stop codon (p.R1087Gfs*12). This alteration occurs at the 3' terminus of the gene, is not expected to trigger nonsense-mediated mRNA decay, and impacts the last 72% of the protein. However, premature stop codons are typically deleterious in nature and a significant portion of the protein is affected (Ambry internal data). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). As such, this alteration is interpreted as a disease-causing mutation.
