Uncertain significance — the classification assigned by Ambry Genetics to NM_006648.4(WNK2):c.6011G>A (p.Ser2004Asn), citing Ambry Variant Classification Scheme 2023: The p.S2004N variant (also known as c.6011G>A), located in coding exon 24 of the WNK2 gene, results from a G to A substitution at nucleotide position 6011. The serine at codon 2004 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
