Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001903.5(CTNNA1):c.416T>C (p.Leu139Pro), citing Ambry Variant Classification Scheme 2023: The p.L139P variant (also known as c.416T>C), located in coding exon 3 of the CTNNA1 gene, results from a T to C substitution at nucleotide position 416. The leucine at codon 139 is replaced by proline, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
