Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001999.4(FBN2):c.2404G>C (p.Asp802His), citing Ambry Variant Classification Scheme 2023: The p.D802H variant (also known as c.2404G>C), located in coding exon 18 of the FBN2 gene, results from a G to C substitution at nucleotide position 2404. The aspartic acid at codon 802 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
