Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001042492.3(NF1):c.5721T>A (p.Phe1907Leu), citing Ambry Variant Classification Scheme 2023: The p.F1886L variant (also known as c.5658T>A), located in coding exon 38 of the NF1 gene, results from a T to A substitution at nucleotide position 5658. The phenylalanine at codon 1886 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
