This variant lies in the NF1 gene (transcript NM_001042492.3) at 5 bases into the intron immediately before coding-DNA position 889, deleting one base. Submitter rationale: The c.889-5delT intronic variant, located in intron 8 of the NF1 gene, results from a deletion of one nucleotide within intron 8 of the NF1 gene. This nucleotide position is highly conserved in available vertebrate species. Using two different splice site prediction tools, this alteration is predicted by ESEfinder to abolish the native splice acceptor site, but is predicted to weaken (but not abolish) the efficiency of the native splice acceptor site by BDGP; however, direct evidence is unavailable. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
