NM_001042492.3(NF1):c.8393A>G (p.Asn2798Ser) was classified as Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N2777S variant (also known as c.8330A>G), located in coding exon 57 of the NF1 gene, results from an A to G substitution at nucleotide position 8330. The asparagine at codon 2777 is replaced by serine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
