NM_001042492.3(NF1):c.6800T>C (p.Ile2267Thr) was classified as Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the NF1 gene (transcript NM_001042492.3) at coding-DNA position 6800, where T is replaced by C; at the protein level this means replaces isoleucine at residue 2267 with threonine — a missense variant. Submitter rationale: The p.I2246T variant (also known as c.6737T>C), located in coding exon 44 of the NF1 gene, results from a T to C substitution at nucleotide position 6737. The isoleucine at codon 2246 is replaced by threonine, an amino acid with similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. To date, this alteration has been detected with an allele frequency of approximately 0.001% (greater than 110000 alleles tested) in our clinical cohort. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
