This variant lies in the NBN gene (transcript NM_002485.5) at coding-DNA position 193, where G is replaced by A; at the protein level this means replaces valine at residue 65 with isoleucine — a missense variant. Submitter rationale: Variant summary: NBN c.193G>A (p.Val65Ile) results in a conservative amino acid change located in the Forkhead-associated (FHA) domain (IPR000253) of the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 1.6e-05 in 251276 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.193G>A in individuals affected with Nijmegen Breakage Syndrome and no experimental evidence demonstrating its impact on protein function have been reported.
