Single allele was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.2760C>G (p.D920E) alteration is located in exon 21 (coding exon 21) of the GIGYF1 gene. This alteration results from a C to G substitution at nucleotide position 2760, causing the aspartic acid (D) at amino acid position 920 to be replaced by a glutamic acid (E). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
