Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001048174.2(MUTYH):c.1111G>A (p.Ala371Thr), citing Ambry Variant Classification Scheme 2023: The p.A399T variant (also known as c.1195G>A), located in coding exon 13 of the MUTYH gene, results from a G to A substitution at nucleotide position 1195. The alanine at codon 399 is replaced by threonine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
