NM_004985.5(KRAS):c.191_196delinsCCAGTT (p.Tyr64_Ala66delinsSerSerSer) was classified as Likely pathogenic for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.191_196delACAGTGinsCCAGTT (p.Y64_A66delinsSSS) alteration, located in exon 3 (coding exon 2) of the KRAS gene, results from an in-frame deletion of ACAGTG and insertion of CCAGTT at nucleotide positions 191 to 196, causing the tyrosine (Y) at amino acid position 64 to be replaced by a serine (S) and the alanine (A) at amino acid position 66 to be replaced by a serine (S). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). These amino acid positions are highly conserved in available vertebrate species. Based on internal structural analysis, this variant is anticipated to disrupt a region of known function (Lu, 2018; Kano, 2019; Pantsar, 2020; Ambry internal data). This alteration is predicted to be deleterious by in silico analysis (Choi, 2012). Based on the available evidence, this alteration is classified as likely pathogenic.
