This variant lies in the CDH2 gene (transcript NM_001792.5) at the canonical splice donor site of the intron immediately after coding-DNA position 1741, where G is replaced by T; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: The c.1741+1G>T intronic variant consists of a G to T substitution one nucleotide after exon 11 (coding exon 11) of the CDH2 gene. Alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay. However, loss of function of CDH2 has not been established as a mechanism of disease. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site.
