Single allele was classified as Pathogenic by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.661delCinsAGCAG (p.R221Sfs*13) alteration, located in exon 7 (coding exon 7) of the GIGYF1 gene, consists of an deletion of 1 and insertion of 5 nucleotides causing a translational frameshift at position 661 with a predicted alternate stop codon after 13 amino acids. This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). Based on the available evidence, this alteration is classified as pathogenic.
