Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000251.3(MSH2):c.331G>A (p.Ala111Thr), citing Ambry Variant Classification Scheme 2023: The p.A111T variant (also known as c.331G>A), located in coding exon 2 of the MSH2 gene, results from a G to A substitution at nucleotide position 331. The alanine at codon 111 is replaced by threonine, an amino acid with similar properties. This amino acid position is highly conserved through mammals but not in all available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
