Uncertain significance for Hereditary nonpolyposis colorectal neoplasms — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000251.3(MSH2):c.988C>A (p.Leu330Met), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the MSH2 gene (transcript NM_000251.3) at coding-DNA position 988, where C is replaced by A; at the protein level this means replaces leucine at residue 330 with methionine — a missense variant. Submitter rationale: This sequence change replaces leucine, which is neutral and non-polar, with methionine, which is neutral and non-polar, at codon 330 of the MSH2 protein (p.Leu330Met). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Experimental studies have shown that this missense change does not substantially affect MSH2 function (PMID: 33357406). Advanced modeling performed at Invitae incorporating data from internal and/or published experimental studies (PMID: 33357406) indicates that this missense variant is not expected to disrupt MSH2 function. ClinVar contains an entry for this variant (Variation ID: 483757). This variant has not been reported in the literature in individuals affected with MSH2-related conditions. This variant is not present in population databases (gnomAD no frequency).
